1. Place a wet tissue at the bottom of the slide box to create a humidified environment. Carefully remove the slides with tissue sections from the cryostat and place them in the slide box (6 slides on each side) or in a wet box. Ensure that the slides do not come into contact with each other.

2. While the slide is still moist, add PBS buffer to the tissue section without spilling it onto the slide.

3. Incubate the slides in PBS containing 0.25% hydrogen peroxide for 30 minutes at room temperature, followed by three washes with PBS.

4. Dilute the primary antibody at 4°C using a microcentrifuge at 13,500 g for 2 minutes. Use 40–50 µl of antibody per slide to cover the tissue section effectively.

5. Using a Pasteur pipette connected to a vacuum pump, gently blot the PBS from one end of the slide and add the primary antibody from the opposite end. Cover the slide with a humidification box and incubate for 1 hour at room temperature.

6. Wash the slide three times with PBS, 5 minutes each time. Add fresh PBS to one end and aspirate the old buffer from the other end.

7. Apply a specific bridged secondary antibody to the slide and incubate for 1 hour at room temperature. Perform three additional PBS washes.

8. Add the PAP complex and let it sit for 1 hour at room temperature. Then, wash the slide three times with PBS.

9. Develop the DAB substrate solution for 2–5 minutes at room temperature. The duration of color development may vary depending on your experimental experience and desired staining intensity.

10. After staining, carefully place the tissue sections back into the slide box and store them in a dark, cool place—avoid freezing to maintain sample integrity.
